Der Ausschuss der Regionen besteht aus 350 Mitgliedern, die die regionalen und lokalen Gebietskörperschaften der 28 Mitgliedstaaten der Europäischen Union vertreten. Er gibt in den gemäß den Verträgen vorgesehenen Fällen obligatorischer Konsultation, fakultativer Konsultation und in eigener Initiative, wenn er dies für zweckdienlich erachtet, eine Stellungnahme ab. Seine Mitglieder sind an keine Weisungen gebunden. Sie üben ihre Tätigkeit unabhängig zum allgemeinen Wohl der Union aus.
