Reģionu komiteju veido 350 locekļi, kuri pārstāv Eiropas Savienības 28 dalībvalstu reģionālās un vietējās pašvaldības. Komiteja sniedz atzinumus obligātas apspriešanās (ja tā ir prasīta Līgumos) un fakultatīvas apspriešanās gadījumos un pēc savas iniciatīvas, ja tā to uzskata par nepieciešamu. Tās locekļiem nav saistoši nekādi obligāti norādījumi. Viņi savus pienākumus veic neatkarīgi, ievērojot Eiropas Savienības vispārējās intereses.
